To understand its action, one must first understand the process of oxidative phosphorylation. In normal cellular respiration, the electron transport chain (ETC) pumps protons across the mitochondrial inner membrane, creating a proton gradient. This gradient drives the enzyme ATP synthase to produce adenosine triphosphate (ATP), the cell's primary energy currency. Mitochondrial uncouplers, like BAM15, disrupt this process by shuttling protons back across the membrane, bypassing ATP synthase. This "shortcircuit" dissipates the proton gradient as heat, thereby reducing the efficiency of ATP production. Consequently, the cell must increase its metabolic rate and oxidize more substrates (including fats and glucose) to meet its energy demands.
